The purpose of this study was to evaluate the performance of the ForenSeq MainstAY kit on the MiSeq FGx system for DNA profiling of forensic challenging samples typically associated with violent crimes, such as firearms and fired cartridge casings. The ForenSeq MainstAY kit targets and sequences 53 forensically informative loci, including 27 autosomal STRs, 25 Y-STRs and the Amelogenin sex marker, meeting both CODIS and ESS requirements. The sequencing results were compared to those obtained through a length-based method using the Investigator 24plex QS kit and amplicon separation through capillary electrophoresis. A complementary objective of the study was to optimize the initial recovery of genetic material for subsequent analysis through an enhanced collection method in order to increase the chances of obtaining a usable DNA profile from trace or highly degraded DNA samples.
Lo scopo del presente studio è stato valutare le prestazioni del kit ForenSeq MainstAY sul sistema MiSeq FGx ai fini del DNA profiling a partire da campioni forensi difficili, tipicamente associati a crimini violenti, come tamponature effettuate su armi da fuoco e bossoli esplosi. Il kit ForenSeq MainstAY sequenzia in un'unica reazione 53 loci informativi per la genetica forense, includendo 27 STR autosomici, 25 Y-STR più il marcatore Amelogenina per la determinazione del sesso, soddisfacendo sia i requisiti del CODIS che dell’ESS. I risultati ottenuti dal sequenziamento sono stati confrontati con quelli ottenuti tramite il metodo di analisi STR basato sulla dimensione degli ampliconi, utilizzando il kit Investigator 24plex QS e la successiva separazione mediante elettroforesi capillare. Obiettivo parallelo dello studio è stato ottimizzare il recupero iniziale del DNA attraverso un pretrattamento dei campioni, al fine di aumentare la probabilità di ottenere un profilo utilizzabile anche a partire da materiale genetico in traccia o altamente degradato.
