Hyperscanning, which measures the temporal alignment between neural activity of individuals interacting with each other as Inter Brain Synchrony (IBS), is an emerging field of neuroscience aimed at uncovering shared patterns of brain activity. Thanks to its intrinsic advantages, such as portability and tolerance of movements, functional near-infrared spectroscopy (fNIRS) represents the most suitable technique for hyperscanning studies in ecological settings. However, given the young age of this field, there are no available guidelines on the optimal pre-processing of fNIRS data yet, and there are no studies investigating the impact of motion correction and physiological noise reduction techniques on the computation of IBS. The aim of this work is to evaluate the impact of different motion correction approaches on inter brain coherence assessment, as well as the effect of systemic components reduction on a dataset collected on two parents (the mother and the father) during the eleven months after the birth of their first child. It emerged that the choice of the most suitable correction method depends on the selected frequency band for the computation of IBS and on the motion artifact type and quantity. Moreover, the impact of physiological reduction techniques on coherence was investigated, by applying either a regression with the use of short-separation channels or the Principal Component Analysis (PCA) technique: the application of these algorithms greatly influenced coherence measurements, possibly leading to ambiguous or incorrect interpretation of the results. Future studies should develop a more controlled, semi-simulated dataset, with a clear ground-truth, to gain a more quantitative evaluation of the most reliable motion correction technique. Furthermore, more robust techniques for physiological noise reduction should be adopted to analyze the nature of the observed discrepancy in coherence before and after correcting physiological noise with the use of PCA. Results shown in this thesis point to an interplay between the choice of the motion correction technique and of the physiological noise reduction technique, with a different impact of physiological noise correction based on the previously applied motion correction technique. Further studies should investigate this interaction to optimize pre-processing guidelines for IBS studies.
L'hyperscanning, che misura l'allineamento temporale tra l'attività neurale di individui che interagiscono tra loro sotto forma di Inter-Brain Synchrony (IBS), è un campo emergente delle neuroscienze che mira a scoprire pattern condivisi di attività cerebrale. Grazie ai suoi vantaggi intrinseci, come la portabilità e la tolleranza ai movimenti, la spettroscopia funzionale nel vicino infrarosso (fNIRS) rappresenta la tecnica più adatta per gli studi di hyperscanning in contesti ecologici. Tuttavia, data la novità di questo campo, non sono ancora disponibili linee guida sul preprocessing ottimale dei dati fNIRS, e non esistono studi che indichino l'impatto delle tecniche di correzione degli artefatti e di riduzione del rumore fisiologico sul calcolo dell'IBS. Lo scopo di questo lavoro è valutare l'impatto di diversi approcci di correzione del movimento sulla valutazione della coerenza inter-brain, nonché l'effetto della riduzione delle componenti sistemiche, su un dataset raccolto su due genitori (la madre e il padre) durante gli undici mesi successivi alla nascita del loro primo figlio. Dallo studio è emerso che la scelta del metodo di correzione più idoneo dipende dalla banda di frequenza selezionata per il calcolo dell'IBS e dal tipo e dalla quantità di artefatti da movimento. Inoltre, è stato studiato l'impatto delle tecniche di riduzione fisiologica sulla coerenza, applicando una regressione con l'uso di canali short-separation oppure la tecnica dell'Analisi delle Componenti Principali (PCA): l'utilizzo di questi algoritmi ha influenzato notevolmente le misure di coerenza, portando potenzialmente a interpretazioni ambigue o errate dei risultati. Studi futuri dovrebbero sviluppare un set di dati più controllato e semi-simulato, con una ground-truth affidabile, per ottenere una valutazione quantitativa della tecnica di correzione degli artefatti più affidabile. Inoltre, dovrebbero essere adottate tecniche più robuste per la riduzione del rumore fisiologico al fine di analizzare la natura della discrepanza osservata nella coerenza prima e dopo la correzione del rumore fisiologico con l'uso della PCA. I risultati mostrati in questa tesi evidenziano un'interazione tra la scelta della tecnica di correzione del movimento e quella di riduzione del rumore fisiologico, con un impatto differente della rimozione delle componenti fisiologiche in base al metodo di riduzione degli artefatti precedentemente applicato. Ulteriori studi dovrebbero approfondire questa interazione per ottimizzare le linee guida di preprocessing negli studi sull'IBS.
