This project is based on photocontrolled degradation of poly-disulfide nanoparticles using a photoswitch called spirothiopyran. Actually , those nanoparticles NPs, already present in literature, consist of a crosslinked α-lipoic acid core and their surface is covered of Pluronic-127 that makes them stealth. Spirothiopyran is an aromatic eterocyclic molecule that, when photoirradiated, isomerizes to a merocyanine, that is a thiolate that can act as glutathione does in biological environment, setting-off depolimerization through nucleophilic attack at disulphide bonds in the crosslinked structure. During this iternship, spirothiopyran was synthesized and characterized with NMR spectroscopy. Then, it was studied how to optimize its photoisomerization and stabilize merocyanine form. Lastly, nanoparticles were endowed with spirothiopyran and it was tried to trigger depolymerization through irradiation with UV-Vis light.
Questo progetto si basa sulla degradazione fotocontrollata di nanoparticelle di polidisolfuro utilizzando un photoswitch chiamato spirotiopirano. In realtà, queste nanoparticelle, già presenti in letteratura, sono costituite da un nucleo di acido α-lipoico reticolato e la loro superficie è ricoperta di Pluronic-127, il che le rende stealth. Lo spirotiopirano è una molecola eterociclica aromatica che, quando fotoirradiata, isomerizza in una merocianina, cioè un tiolato che può agire come fa il glutatione in ambiente biologico, innescando la depolimerizzazione attraverso l'attacco nucleofilo ai legami disolfuro nella struttura reticolata. Durante questo tirocinio, lo spirotiopirano è stato sintetizzato e caratterizzato mediante spettroscopia NMR. Successivamente è stato studiato come ottimizzarne la fotoisomerizzazione e stabilizzare la forma della merocianina. Infine, lo spirotiopirano è stato incapsulato dentro le nanoparticelle e si è cercato di innescare la depolimerizzazione attraverso irradiazione con radiazione elettromagnetica UV-Visibile.
