Abstract

The discovery of the TrxR2 inhibition by Auranofin, for a long time only used as an antiarthritic drug, led to extensive research focused on the utilisation of gold(I) complexes as anticancer agents. The relevance of this discovery lies in the key role played by Thioredoxin reductase, as it acts as a redox regulator within the cells and it is thus implicated in the tumour progression; the ability of gold(I) complexes to inhibit this enzyme by altering the redox homeostasis gives interesting perspectives on the use of gold in the oncological field. Suntharalingam and coworkers, in a study published in 2023, reported the promising activity of the compound [Au(PPh3)indomethacin] against breast cancer stem cells. Basing on these results, Scattolin and coworkers drew attention to [Au(NHC)(indomethacin)] complexes, which exhibited a notable antiproliferative effect on various cell lines, some of them resistant to cisplatin.
