The present study aims at the development and characterization of novel antimicrobial peptides (AMPs) and their conjugation onto cotton for the development of medical textiles. Starting from two model peptides already known for their antibacterial activity (peptides 3.0 and 3.1), two structural analogues were designed and synthesized de novo. These analogues feature 2-aminoisobutyric acid and/or a palmitic derivative in their sequence in order to enhance antimicrobial activity and resistance to enzymatic degradation. The most promising peptides were characterized using circular dichroism and enzymatic stability assays, while their antimicrobial efficacy was tested against S. aureus, E. coli, and A. baumannii. The results show that the introduction of the non-proteinogenic amino acid leads to lower MIC values for the three bacterial strains and to increased stability. The final phase of the project involved the covalent anchoring of the best candidate onto modified cotton, using an approach based on oxime formation, in order to obtain a bioactive material.
