Monoclonal antibodies make up roughly half of all approved biologics on the market. Being proteins however, they are susceptible to oxidation during either the manufacturing process or the storage conditions, which can occur via several pathways, such as metal-catalyzed oxidation, due to the presence of redox-active metals, photooxidation, due to exposure to light, or the presence/formation of reactive oxygen species (ROS). Oxidation can have a profound effect on therapeutics' stability, immunogenicity and efficacy, therefore having a reliable method to predict oxidation sites and its subsequent effects could prove useful in both drug design and assessment of storage conditions. In this study, Bevacizumab was oxidized with AAPH (2,2'-Azobis(2-amidinopropane) dihydrochloride) and then analyzed with LC-MS/MS, CD-UV spectroscopy and UV spectroscopy thus obtaining experimental information about the oxidized residues and effects on the structure; in-silico methods were then used to validate the existing oxidation prediction models and to characterise the effects at a molecular level.

Abstract
In this study, Bevacizumab was oxidized with AAPH (2,2'-Azobis(2-amidinopropane) dihydrochloride) and then analyzed with LC-MS/MS, CD-UV spectroscopy and UV spectroscopy thus obtaining experimental information about the oxidized residues and effects on the structure; in-silico methods were then used to validate the existing oxidation prediction models and to characterise the effects at a molecular level.| File | Dimensione | Formato | |
