In this Thesis the synthesis, chemical characterization and conformational analysis in solution of two series of peptides containing Cα-tetrasubstituted amino acids that generate an ordered helical structure of great stability, that may be used to calibrate new spectroscopic techniques. These peptides were prepared using total solution synthesis, both “step by step” and “fragment condensation” approach. Conformational analysis in solution (IR and EPR) confirmed that these peptides adopt a helical conformation, primarily 310- helical type. In particular, they will be studied by high frequency (95 GHz) EPR, using PELDOR technique (Prof. Martina Huber, Leiden, The Netherlands).
