The goal of this thesis work was firstly the characterisation (i.e. the study of the acidic dissociation constants) of two cyclen-based chelators; one of these chelators was synthesized at the British Columbia University of Vancouver and the other one at the University of Padua. After the determination of the chelators’ acidic dissociation constants, the goal of the following measurements was the kinetic and thermodynamic study of systems formed by these chelators with Cu(II) and Pb(II), which are borderline metal ions of radiopharmaceutical interest, in order to determine the number, stoichiometry, and formation constants of the complexes and the rate of the complexation reaction between metal and chelator. The measurements were carried out by potentiometry, UV-vis spectrophotometry and NMR spectroscopy.
Lo scopo di questo lavoro di tesi è stato in primo luogo la caratterizzazione, intesa come studio delle costanti di dissociazione acida, di due molecole chelanti aventi in comune la struttura base di tipo cyclen; una di queste molecole è stata sintetizzata presso la British Columbia University di Vancouver, mentre l’altra presso l’Università degli Studi di Padova. Dopo la determinazione delle costanti di dissociazione acida dei leganti, le misure successive hanno avuto come obiettivo lo studio cinetico e termodinamico di sistemi formati dai leganti suddetti con Cu(II) e Pb(II), ioni metallici borderline di interesse radiofarmaceutico, al fine di determinare il numero, la stechiometria, le costanti di complessamento dei complessi formati e la velocità della reazione di complessamento tra metallo e legante. Le misure sono state condotte mediante potenziometria, spettrofotometria UV-vis e spettroscopia NMR.
