The purpose of the study is to investigate the presence of transgenerational effects of two different PFASs in Poecilia reticulata. The PFASs used are GenX (ammonium 2,3,3,3-tetrafluoro-2-(heptafluoropropoxy)-propanoate) and PFOA (perfluorooctanoic acid), and because the objective was to study transgenerational effects, the fish exposed to PFASs are not the ones under study, but their parents. With a control group, born to parents not exposed to PFAS, it was possible to study the differences between the groups through morphological measurements, fertility, behavioral tests, and resistance tests. Regarding morphological measurements, standard length was measured at 1 day old, after 2 months and after 5 months. For 1-day-old fry, brain area and width were also analyzed, while for 5-month-old adult males, spot area was also analyzed. The fertility of females was then assessed by collecting their fry, and the quality of male ejaculate was also studied. A resistance test was then carried out using an instrument called a flow chamber. Finally, behavioral tests were used to study the boldness of the fish.
Lo scopo dello studio è indagare la presenza di effetti transgenerazionali di due diversi PFAS in Poecilia reticulata. I PFAS usati sono GenX (2,3,3,3-tetrafluoro-2-(eptafluoropropossi)-propanoato di ammonio) e PFOA (Acido perfluoroottanoico) e, poichè l’obbiettivo era studiare gli effetti transgenerazionali, i pesci esposti ai PFAS non sono quelli oggetto dello studio, ma i loro genitori. Grazie ad un gruppo di controllo, nato da genitori non esposti a PFAS, è stato possibile studiare le differenze tra i diversi gruppi attraverso misurazioni morfologiche, di fertilità, test comportamentali e di resistenza. Per quanto riguarda le misurazioni morfologiche, è stata misurata la lunghezza standard a 1 giorno di vita, dopo 2 mesi e dopo 5 mesi. Per gli avannotti di un giorno è stata analizzata anche l’area del cervello e la larghezza, mentre per i maschi adulti di 5 mesi è stata analizzata anche l’area delle macchie. È poi stata valutata la fertilità delle femmine raccogliendone gli avannotti ed è anche stata studiata la qualità dell’eiaculato maschile. È stato poi effettuato un test di resistenza tramite uno strumento chiamato flow chamber. Infine, dei test comportamentali hanno permesso di studiare la boldness dei pesci.
