Deinococcus radiodurans is a polyextremophile with the remarkable ability to survive various types of environmental stress, particularly ionizing and UV radiation. This thesis analyses and discusses in detail a study examining the heterologous expression in Escherichia coli of the Deinococcus radiodurans genes RecA and uvrD, and of the Dsup gene from the tardigrade Ramazzottius varieornatus, with the aim of enhancing the host's resistance to various environmental stresses. Expression of the deinococcal genes did not increase the survival rate, likely due to the incompatibility between them and the DNA repair machinery of E. coli. Expression of Dsup, by contrast, increased the survival rate, supporting its independent role in the physical protection of DNA. The radioresistance mechanisms of D. radiodurans continue to be the subject of numerous studies aimed at engineering microbial radioresistance, particularly in the fields of astrobiology and bioremediation.
Deinococcus radiodurans è un poliestremofilo che possiede la straordinaria capacità di sopravvivere ai vari tipi di stress ambientali, soprattutto alle radiazioni ionizzanti e UV. In questa tesi viene analizzato e discusso nello specifico uno studio che prende in esame l’espressione eterologa in Escherichia coli dei geni RecA e uvrD di Deinococcus radiodurans e del gene Dsup del tardigrado Ramazzottius varieornatus, con lo scopo di migliorarne la resistenza a vari stress ambientali. L’espressione dei geni deinococcali non ha aumentato il tasso di sopravvivenza, verosimilmente a causa dell’incompatibilità tra essi e il macchinario di riparazione del DNA di E. coli. L’espressione di Dsup invece ha aumentato il tasso di sopravvivenza, a sostegno del suo ruolo indipendente nella protezione fisica del DNA. I meccanismi di radioresistenza di D. radiodurans continuano ad essere oggetto di numerosi studi per l’ingegnerizzazione della radioresistenza microbica, specialmente nell’ambito dell’astrobiologia e del biorisanamento.
