During the course of my internship for my bachelor's thesis, I conducted a first and preliminary analysis of the male reproductive capability of a KO mutant line of zebrafish, for the mytho gene. This gene is very expressed in the gonads of zebrafish and it is therefore possible to suggest that the Mytho protein has a role in reproductive functions. We found a significative reduction in the reproductive success of males. So, we decided to focus on the study of possible morphological alterations. We analyzed morphology with both optical and electronic microscopy, and found, thanks to the latter, an alteration in nuclei shape of sperm cells and an increased number of autophagic structures in the mutant testicles, compared to the wild-type (WT). Finally, an histological analysis with the optical microscope revealed the presence of at least two individuals with testicular seminomas, a TGCT.
Durante il mio internato di tesi triennale mi sono occupata di condurre una prima e preliminare analisi delle capacità riproduttive maschili della linea knock-out (KO) di zebrafish per il gene mytho. Questo gene risulta essere molto espresso nelle gonadi dello zebrafish e quindi è possibile ipotizzare che la proteina Mytho possa avere un ruolo in ambito riproduttivo. La riduzione delle capacità riproduttive è risultata significativa e quindi ci siamo focalizzati sullo studio di possibili alterazioni morfologiche. La morfologia è stata analizzata sia a livello di microscopia ottica che elettronica, evidenziando, in quest’ultimo caso, l’alterazione della forma del nucleo dello sperma e la presenza di un numero superiore di figure autofagiche nei testicoli dei mutanti rispetto ai wild-type (WT). Infine, l’analisi istologica al microscopio ottico ha portato al rilevamento di almeno due esemplari di seminomi testicolari, una comune forma di TGCT.
