Deslorelin is a gonadotropin-releasing hormone (GnRH) agonist licensed for pharmacological sterilization in adult male dogs and cats, prepubertal bitches, and used off-label in queens. Its efficacy and safety for reproduction control in female cats has been demonstrated, however its routine clinical use remains limited. This study aims to evaluate the long-term effects of 9.4 mg deslorelin treatment on the immune system of post-pubertal female cats, incorporating and expanding on findings from a previous study on males. Nine post-puberal intact queens were enrolled in the study based on strict health criteria: vaccinated, dewormed, unremarkable physical and reproductive examination and bloodwork, and absence of previous reproductive disorders. Oral and written informed consents were obtained from the owners before the beginning of the study procedures. Each cat received a 9.4 mg deslorelin implant (Suprelorin®, Virbac) and was monitored through monthly owner interviews and biannual veterinary appointments, where blood was collected for hemogram, biochemistry, progesterone and cytokine assays. Nineteen cytokines were assayed at T0, T1 and T2 (pre-implantation, 6 and 12 months post-implantation, respectively) in plasma and peripheral blood mononuclear cells (PBMCs) culture. The cytokines were assayed using a multiplex feline-specific cytokine immunoassay kit. A repeated mixed model ANOVA was applied to compare cytokine levels throughout time (from T0 to T2) in female and male cats both in plasma and PBMCs. The treatment was effective in all subjects, and by this time seven out of eight queens have reached the end of the implant’s effect, which lasted from 648 up to 988 days. One cat died due to unrelated causes while still under the effect of the implant at day 601, and for the remaining subject the implant remains effective beyond two years. Three cytokines showed significant plasma-level modifications in females: TNF-α and IL-1β increased significantly, while SCF initially increased and then stabilised. In male cats, one cytokine (MCP) increased in plasma T1-T2. The overall analysis done unifying queens and tomcats’ groups revealed that five cytokines had significant changes throughout time in PBMC culture (FLT-3L, FAS, IL-4, IL-2, KC). Notably, FLT-3L and IL-1β exhibited a distinct response between males and females, in PBMC and in plasma analysis, respectively. The implant duration was coherent with results of other studies, and the implant’s safety was confirmed. In the present study, four cytokines (TNF- α, IL-1β, IL-2, KC) showed significant increase following deslorelin implantation from T0 to T2, the former two only in the female population and the latter two in the overall population of females and males. Additionally, two cytokines (SCF, FAS) initially changed between T0-T1 and then stabilized. Moreover, three cytokines (MCP, FLT-3L, IL-4) exhibited a delayed rise between T1-T2. In the overall group, one cytokine (KC) showed a different increase throughout timepoints, more marked in tomcats. It is possible that sex plays a role in the onset of immunomodulation triggered by deslorelin. Notably, the sex-related changes showed by two cytokines (IL-1β, FLT-3L) became significant combining the groups for the statistical analysis. The underlying mechanisms remain unclear, however it seems that cytokine changes reflected an adaptive modulation of the immune system associated with the reproductive modifications induced by the implant, rather than immunocompromise derived from the treatment. This study is the first to focus on the potential effects on the immune system measuring cytokines after a deslorelin implantation in cats. Further research is needed to elucidate the full impact of deslorelin on feline immune function and to rule out a negative effect on the immune system.

Il deslorelin è un agonista dell’ormone di rilascio delle gonadotropine (GnRH) il cui uso nelle gatte è off-label. Sebbene efficacia e sicurezza nel controllo della riproduzione nella gatta siano state dimostrate, il suo impiego clinico rimane limitato. Questo studio valuta gli effetti a lungo termine sul sistema immunitario di un trattamento con deslorelin 9.4 mg di gatte post-puberi, ampliando i risultati di un precedente studio nei maschi. Nove gatte intere post-puberi sono state incluse nello studio secondo rigorosi criteri sanitari: vaccinazioni e trattamenti antiparassitari regolari, visita clinica e riproduttiva nella norma, esami ematochimici nella norma e assenza di pregresse patologie riproduttive. I proprietari hanno fornito consenso informato orale e scritto. Ogni soggetto ha ricevuto un impianto di deslorelin da 9.4 mg (Suprelorin®, Virbac) ed è stato monitorato mediante chiamate mensili ai proprietari e visite veterinarie semestrali, durante le quali venivano raccolti campioni ematici per emocromo, biochimico, progesterone e analisi citochiniche. Diciannove citochine sono state dosate a T0, T1 e T2 (pre-impianto, 6 e 12 mesi post-impianto rispettivamente) sia nel plasma sia in colture di cellule mononucleate del sangue periferico (PBMCs), utilizzando un kit immunologico multiplex specifico per il gatto. Per confrontare i livelli citochinici nel tempo è stata applicata un’ANOVA a misure ripetute con modello misto. Il trattamento si è dimostrato efficace in tutti i soggetti. Sette gatte su otto hanno concluso l’effetto dell’impianto, con una durata compresa tra 648 e 988 giorni. Una gatta è deceduta per cause non correlate mentre l’impianto era ancora efficace, mentre nell’ultimo soggetto l’effetto persiste oltre i due anni. Nelle femmine, tre citochine hanno mostrato modificazioni significative a livello plasmatico: TNF-α e IL-1β sono aumentate significativamente, mentre SCF ha mostrato un incremento iniziale seguito da stabilizzazione. Nei maschi, solo la citochina MCP nel plasma ha mostrato un cambiamento significativo tra T1 e T2. L’analisi complessiva dei gruppi maschili e femminili ha evidenziato variazioni significative nel tempo per cinque citochine nelle PBMCs (KC, IL-4, FLT-3L, FAS e IL-2). FLT-3L e IL-1β hanno inoltre mostrato una risposta differente tra maschi e femmine rispettivamente nelle PBMCs e nel plasma. La durata dell'impianto è risultata coerente con quella riportata in altri studi, e la sicurezza dell’impianto è stata confermata. Sono necessarie ulteriori ricerche per chiarire l’impatto del deslorelin sulla funzione immunitaria felina e garantirne un impiego sicuro nella pratica clinica.
