The search for effective alternatives to platinum-based anticancer drugs represents one of the most significant challenges in bioinorganic and pharmaceutical chemistry. In this context, cobalt complexes have attracted growing scientific interest due to their unique redox properties and ability to be selectively activated in the tumor microenvironment. This thesis analyzes the main cobalt complexes with potential anticancer activity reported in the literature, with particular attention to their chemical, physical, and structural properties, mechanisms of interaction with biological targets, and cytotoxic activity data obtained in in vitro and in vivo studies. The aim is to provide an updated overview of the therapeutic potential of these complexes, highlighting their development prospects as new metallodrugs for the treatment of cancer.
La ricerca di alternative efficaci ai farmaci antitumorali a base di platino rappresenta una delle sfide più rilevanti nell'ambito della chimica bioinorganica e farmaceutica. In questo contesto, i complessi di cobalto hanno suscitato un crescente interesse scientifico grazie alle loro peculiari proprietà redox e alla capacità di essere attivati selettivamente nel microambiente tumorale. Il presente lavoro di tesi analizza i principali complessi di cobalto a potenziale attività antitumorale riportati in letteratura, con particolare attenzione alle loro proprietà chimico-fisiche e strutturali, ai meccanismi di interazione con i bersagli biologici e ai dati di attività citotossica ottenuti negli studi in vitro e in vivo. L'obiettivo è fornire un quadro aggiornato delle potenzialità terapeutiche di questi complessi, evidenziandone le prospettive di sviluppo come nuovi metallofarmaci per il trattamento delle neoplasie.
