Functionalization with polyethylene glycol (PEG) provides nanoparticles with greater stability, but this also hinders cellular internalization. One solution to this problem is to develop systems capable of losing PEG only upon reaching the target. This thesis proposes the design of gold nanoparticles noncovalently functionalized with specifically functionalized PEG chains. The system's architecture is designed so that, in the presence of specific concentrations of ATP, the nanoparticles lose their PEG coating, thus facilitating their cellular uptake. This thesis synthesized key intermediates for the synthesis of the PEG-based derivative.
La funzionalizzazione con polietilenglicole (PEG) garantisce alle nanoparticelle una maggior stabilità alle nanoparticelle, ma il questo ha anche l'effetto di ostacolare l'internalizzazione cellulare. Per risolvere questo problema, una soluzione può essere quella di sviluppare sistemi in grado di perdere il PEG solo una volta arrivati in prossimità del bersaglio. Questo lavoro di tesi propone la progettazione di nanoparticelle d'oro funzionalizzate in modo non covalente con catene di PEG appositamente funzionalizzate. L'architettura del sistema è pensata affinché in presenza di determinate concentrazioni di ATP, le nanoparticelle perdano il PEG che le ricopre, in modo da facilitarne l'assorbimento cellulare. Durante questo lavoro di tesi sono stati sintetizzati degli intermedi chiave per la sintesi del derivato a base di PEG.
