Abstract

CRISPR-Cas9 system-mediated genome editing represents a fundamental biotechnological innovation resulting from the conversion of a bacterial immune system. Operating as a “molecular scissors”, the Cas9 endonuclease creates specific cuts on target DNA, triggering natural repair mechanisms. This paper analyzes the molecular architecture of the system, exploring its protein domains and specificity of action. The theoretical foundations are subsequently applied to the characterization of the ATAD3B gene, encoding a mitochondrial protein which, at the laboratory where this thesis was carried out, was identified as selectively expressed in some types of highly malignant and metastatic tumors. For this reason, our laboratory hypothesizes that its silencing may determine, at least partially, a reversion of the malignant phenotype associated with it. Due to the very high sequence homology (greater than ’85%) to the paralog gene ATAD3A, traditional gene manipulation techniques frequently cause nonspecific silencing and cell mortality. The use of CRISPR-Cas9 technology allows, on the contrary, to induce a targeted and highly specific knockout on ATAD3B, with the ultimate goal of defining the function and thoroughly investigating its pathophysiological role.
