The Enzyme-linked Immunosorbent Assay (ELISA) is an effective biochemical assay for the detection and quantification of proteins and peptides. It is based on the use of antibodies that interact specifically with the target molecule and are conjugated with enzymes whose interaction with a substrate produces a chromophore. Abstract

By measuring the absorbance of the chromophore, the target molecule can also be quantified. In this thesis the various types of ELISA and their principle of operation will be discussed. In particular, the ELISA protocol that has been applied in the laboratory to measure the concentration of different cytokines produced by Tumor Associated Macrophages (TAM) will be explored, describing the materials used, its steps in detail and the obtained results.
