MicroRNAs (miRNAs) are a class of noncoding RNAs that regulate gene expression. Among them, miR-210 is one of the most evolutionarily conserved and it’s involved in numerous physiological and pathological processes, largely in response to hypoxic conditions. It has recently been observed that miR-210 knockout can lead to retinal degeneration in both Drosophila melanogaster and Mus musculus. To investigate and understand the importance of miR-210 in ocular tissues, this thesis evaluated its expression profiles in cellular models of retinal differentiation and in human corneal tissues. After extracting total RNA and assessing its purity and integrity, we then performed quantitative PCR (qRT-PCR). The results of this analysis highlighted the expression of miR-210 in all analyzed samples, with different expression levels both during the developmental stages of the retinal pigment epithelium and in the corneas of different patients. The observed variability in expression levels suggests a possible involvement of this gene in the development and maintenance of ocular homeostasis in humans as well. However, this role will need to be studied in deeper through further studies.
