In viticulture Plasmopara viticola, the causative agent of downy mildew of the vine, represents one of the main pathogens, as most of the varieties of Vitis vinifera grown today are susceptible to it. The predominant form of defense against the pathogen is represented by copper which, in organic farms, is the only permitted instrument of struggle. However, the limitations regarding the use of copper are increasingly severe and stringent, since, being a heavy metal poorly absorbed by plant tissues, it tends to accumulate in the soil and become toxic to the environment, animals and humans. It is therefore essential to seek alternative strategies that effectively counteract the pathogen while reducing the use of copper. In this regard, in the last twenty years the study of nanotechnologies for the management of diseases in agriculture has had a strong increase. Given the following premises, the aim of this paper was to evaluate the efficacy against Plasmopara viticola, the persistence on the leaf blade following leaching and the phytotoxicity of copper nanoparticles (CuO NPs), in comparison with a commercial product. based on copper sulphate (TRICOPPERLAND) with a copper metal concentration of 48 mg / 100 ml, as per label. Initially, the dosage of copper present on leaves treated in the field was analyzed to define the optimal dose to be distributed in laboratory tests and at the same time construct a decay curve of the copper itself in the following days. Two different methods, spectrophotometry and X-Ray Fluorescence (XRF), were used and compared to quantify the copper on the harvested leaves. The quantity of copper metal to be distributed on the leaves was therefore calculated as 0.001 mg / cm2 of leaf, which corresponds to about 1/5 of the full field dose of the commercial formulation. Once the optimal dose of copper metal was established, the effectiveness of the TRICOPPERLAND treatments dose 1: 5 (9.6 mg / 100 ml), CuO dose 1: 5 (9.6 mg / 100 ml) and CuO was then tested in the laboratory. dose 1:10 (4.8 mg / 100 ml) on grape leaves eleven days after inoculation with P. viticola: although in the treatment with TRICOPPERLAND the infection is slightly less than in both theses treated with CuO nanoparticles , no significant differences were observed between treatments. As for persistence, by applying a washout equal to 1 L / m2 two days after treatment, the CuO nanoparticles at both concentrations were poorly leached (12% and 17% respectively) compared to TRICOPPERLAND (35%). The results therefore show a greater adhesion to the leaf surface of the nanoparticles and a similar control action compared to the copper of the commercial formulation. From the phytotoxicity tests, carried out on vine leaves, it was also seen that the CuO nanoparticles are significantly less phytotoxic than the commercial formulation. The results obtained from the following thesis represent a starting point for future studies on the applications of copper-based nanoformulates in viticulture.

In viticoltura Plasmopara viticola, agente eziologico della peronospora della vite, rappresenta uno dei patogeni principali, in quanto la maggior parte delle varietà di Vitis vinifera ad oggi coltivate ne è suscettibile. La forma preponderante di difesa contro il patogeno è rappresentata dal rame che, nelle aziende biologiche, è l’unico strumento di lotta ammesso. Tuttavia, le limitazioni riguardo l’impiego del rame sono sempre più severe e stringenti, in quanto, essendo un metallo pesante scarsamente assorbito dai tessuti vegetali, tende ad accumularsi nel terreno e a diventare tossico per l’ambiente, gli animali e l’uomo. È dunque fondamentale ricercare strategie alternative che contrastino efficacemente il patogeno riducendo al contempo l’utilizzo di rame. A tal proposito negli ultimi vent’anni ha avuto un forte incremento lo studio delle nanotecnologie per la gestione delle malattie in ambito agricolo. Date le seguenti premesse, l’obiettivo di questo elaborato è stato quello di valutare l’efficacia contro Plasmopara viticola, la persistenza sulla lamina fogliare in seguito a dilavamento e la fitotossicità di nanoparticelle di rame (CuO NPs), a confronto con un prodotto commerciale a base di solfato di rame (TRICOPPERLAND) con concentrazione di rame metallo di 48 mg/100 ml, come da etichetta. Inizialmente è stato analizzato il dosaggio di rame presente su foglie trattate in campo per definire la dose ottimale da distribuire nelle prove di laboratorio e contestualmente costruire una curva di decadimento del rame stesso nei giorni successivi. Per quantificare il rame sulle foglie raccolte sono stati utilizzati e confrontati due differenti metodi, spettrofotometria e X-Ray Fluorescence (XRF). La quantità di rame metallo da distribuire sulle foglie è stata quindi calcolata in 0,001 mg/cm2 di foglia, che corrisponde a circa 1/5 della dose piena di campo del formulato commerciale. Stabilita la dose ottimale di rame metallo, in laboratorio è stata quindi testata l’efficacia dei trattamenti TRICOPPERLAND dose 1:5 (9,6 mg/100 ml), CuO dose 1:5 (9,6 mg/100 ml) e CuO dose 1:10 (4,8 mg/100 ml) su foglie di vite undici giorni dopo l’inoculo con P. viticola: nonostante nel trattamento con TRICOPPERLAND l’infezione risulti essere leggermente minore rispetto ad entrambe le tesi trattate con nanoparticelle di CuO, non si sono osservate differenze significative tra i trattamenti. Per quanto riguarda la persistenza, applicando un dilavamento pari ad 1 L/m2 due giorni dopo il trattamento, le nanoparticelle di CuO ad entrambe le concentrazioni sono state scarsamente dilavate (rispettivamente del 12% e 17%) rispetto al TRICOPPERLAND (35%). I risultati evidenziano, quindi, una maggior adesione alla superficie fogliare delle nanoparticelle ed una simile azione di controllo rispetto al rame del formulato commerciale. Dai saggi di fitotossicità, effettuati su foglie di vite, si è visto inoltre che le nanoparticelle di CuO sono significativamente meno fitotossiche rispetto al formulato commerciale. I risultati ottenuti dalla seguente tesi rappresentano un punto di partenza per studi futuri sulle applicazioni di nanoformulati a base di rame in viticoltura.
