As reported in literature, in a pathological phenotype such as Acute Myeloid Leukemia, there is a high expression of a protein called Adrenomedullin. This hormone is physiologically released in response to stress factor such as hypoxia, resulting in vasodilation and angiogenesis. Regarding tumor microenvironment, it appears to be associated with cell survival mechanisms, resulting in an anti-apoptotic response In this study I tried to characterize the effect induced by ADM and its antagonist peptide ADM22-52 in the control of tumor growth in a leukemia cell line. Particular attention was given to the role of the RAMP3 receptor in the control of leukemic growth.
Studi in letteratura mettono in luce come in un fenotipo patologico quale la Leucemia Mieloide Acuta, vi sia un'alta espressione di una proteina detta Adrenomedullina. Tale ormone fisiologicamente viene rilasciato in risposta a fattori di stress come l’Ipossia, determinando vasodilatazione e Angiogenesi. Per quanto riguarda il microambiente tumorale sembra essere associata a meccanismi di sopravvivenza cellulare, determinando una risposta anti-apoptotica In questo studio mi sono proposto di caratterizzare in una linea cellulare leucemica, l’effetto indotto da ADM e da un suo peptide antagonista ADM22-52 nel controllo della crescita tumorale. Particolare attenzione è stata data al ruolo del recettore RAMP3 nel controllo della crescita leucemica.
