MicroRNAs, molecules involved in gene expression, are frequently deregulated in many human diseases and cancers. In recent years, many studies have shown a deregulation of these molecules in chronic liver diseases and hepatocellular carcinoma, suggesting a possible diagnostic and prognostic role. One of the most expressed microRNAs in chronic liver diseases is miR-21. The aim of the project is to understand the impact of miR-21 modulation on hepatic triglycerides during diabesity. In particular our team purpose is to understand molecular mechanisms underlying the pathogenic role of microRNA-21 in hepatocytes of obese patients. It includes three steps 1) Identify the direct target RNAs of mir21 by using the iCLIP2-seq technique and RNA-seq. 2) Validate in cells the most promising, and those releted to liver metabolism, dysregulation target RNAs of miR21 by several biochemical and molecular techniques, such as: - western Blot - qPCR - Gene Reporter Assay 3) Investigate in cells the physiological and pathological role of miR-21 targets by performing a global bioinformatic analysis, such as IPA analysis.
I microRNA, molecole coinvolte nell'espressione genica, sono frequentemente deregolati in molte malattie e tumori umani. Negli ultimi anni molti studi hanno mostrato una deregolazione di queste molecole nelle malattie croniche del fegato e nel carcinoma epatocellulare, suggerendo un possibile ruolo diagnostico e prognostico. Uno dei microRNA più espressi nelle malattie croniche del fegato è il miR-21. Lo scopo del progetto è comprendere l'impatto della modulazione del miR-21 sui trigliceridi epatici durante la diabesità. In particolare, lo scopo del nostro team è comprendere i meccanismi molecolari alla base del ruolo patogeno del microRNA-21 negli epatociti di pazienti obesi. Comprende tre passaggi 1) Identificare gli RNA target diretti di mir21 utilizzando la tecnica iCLIP2-seq e RNA-seq. 2) Convalidare nelle cellule i più promettenti, e quelli legati al metabolismo epatico, la disregolazione degli RNA bersaglio di miR21 mediante diverse tecniche biochimiche e molecolari, come: - macchia occidentale - qPCR - Saggio del reporter genetico 3) Indagare nelle cellule il ruolo fisiologico e patologico dei bersagli miR-21 eseguendo un'analisi bioinformatica globale, come l'analisi IPA.
