Atherosclerosis is an inflammatory disease that arises as a consequence of the deposition of lipids in the walls of medium and large caliber arteries. Hypercholesterolemia is a metabolic disorder characterized by higher LDL-C and total cholesterol levels. As such, it is a favorable condition for the onset of atherosclerosis and other pathologies. Its high incidence is largely due to sedentary lifestyle, irregular diet or even genetic alterations. This thesis deals with the pharmacological and non-pharmacological therapies that are currently most effective in reducing plasma LDL-C and total cholesterol, as well as reducing the incidence of cardiovascular events and the size of atheromatous plaques. The discussion starts from the standard therapies, whose efficacy and safety has been widely demonstrated over the years, and then continues with the description of the currently used novel therapies.
L’aterosclerosi è una patologia a carattere infiammatorio che insorge in seguito al deposito di lipidi a livello della parete delle arterie di medio e grande calibro. L’ipercolesterolemia è un disordine metabolico caratterizzato da livelli plasmatici di LDL-C e colesterolo totale superiori rispetto alla norma. Come tale, se non trattata, risulta essere una condizione favorevole all’insorgenza di aterosclerosi e di altre patologie. La sua elevata incidenza è dovuta in gran parte allo stile di vita sedentario e alla dieta irregolare dei pazienti, ma anche ad alterazioni genetiche. In questo elaborato sono state trattate le terapie, farmacologiche e non, ad oggi maggiormente efficaci nella riduzione dei valori plasmatici di LDL-C e colesterolo totale, oltre che nella riduzione dell’incidenza di eventi cardiovascolari e della dimensione delle placche ateromatose. La trattazione parte dalle terapie standard, la cui efficacia e sicurezza è stata ampiamente dimostrata nel corso degli anni, per poi proseguire nella descrizione delle nuove terapie attualmente utilizzate.
