Flavescenza dorata in vigneti del Veneto: studio del rischio di infezione derivante da fonti esterne

This thesis will deal with Flavescenza dorata of the grapevine, a disease that is increasingly present in vineyards in the Veneto, Italy and Europe. Flavescenza dorata is a disease that belongs to the group of grapevine yellows and is associated with a phytoplasma belonging to the 16Sr V group, mainly carried in nature by the leafhopper, Scaphoideus titanus. The aim of this thesis is to evaluate, in selected vineyards where the disease is very present, if it is due to the exclusive activity of S. titanus, or also to alternative vectors, which colonise hedges or spontaneous plants bordering the vineyards, and which could move to these. Leaf samples were taken from symptomatic vines. The samples were subjected to DNA extraction, followed by amplification, by Real-Time PCR, direct PCR or nested PCR, of two phytoplasma gene regions: 16S-23Sr DNA and secY-map. The analyses are still in progress, and the data will be processed and discussed.

In questa tesi verrà trattato il tema della Flavescenza dorata della vite, malattia sempre più presente nei vigneti veneti, italiani ed europei. La Flavescenza dorata è una malattia che fa parte del gruppo dei Giallumi della vite ed è associata ad un fitoplasma appartenente al gruppo 16Sr V, veicolato in natura principalmente dalla cicalina, Scaphoideus titanus. Obiettivo della tesi è quello di valutare, su vigneti già selezionati in cui la malattia è molto presente, se questa è dovuta all’esclusiva attività di S. titanus, o anche a vettori alternativi, che colonizzano siepi o piante spontanee limitrofe ai vigneti, e che potrebbero spostarsi su questi. Sono stati prelevati campioni fogliari da viti sintomatiche. I campioni sono stati sottoposti ad estrazione del DNA, seguita da amplificazione, mediante PCR Real-Time, PCR diretta o nested, di due regioni geniche del fitoplasma: 16S-23Sr DNA e secY-map. Le analisi sono ancora in corso, i dati saranno elaborati e discussi.
