My thesis project involved the synthesis of an ADC (Antibody Drug Conjugate) directed against the GPC-1 receptor, overexpressed in pancreatic cancer. I used an IgM as an antibody and MMAE as an anticancer drug. Two different types of conjugates have been produced, exploiting different types of synthesis: lysine conjugates and cysteine conjugates. The first step was the characterization of the IgM and then the actual synthesis and characterization of conjugates obtained through spectroscopic, colorimetric, electrophoretic and chromatographic measurements. The different conjugates were then also tested in vitro, on two different cell lines, to see the actual activity of the conjugates themselves.
Il mio progetto di tesi prevedeva la sintesi di un ADC (Antibody Drug Conjugate) diretto contro il recettore GPC-1, sovraespresso a livello del tumore pancreatico. Ho utilizzato una IgM come anticorpo e MMAE come farmaco antitumorale. Sono stati prodotti due diversi tipi di coniugati, sfruttando tipologie di sintesi diverse: coniugati lisinici e coniugati cisteinici. Il primo step è stata la caratterizzazione dell' IgM per poi passare alla sintesi vera e propria e alla caratterizzazione dei coniugati ottenuti tramite misure spettroscopiche, colorimetriche, elettroforetiche e cromatografiche. I diversi coniugati sono poi stati testati anche in vitro, su due linee cellulari diverse, per vedere l'effettiva attività dei coniugati stessi.
