The Quantitative Real Time PCR (qPCR) technique is a DNA amplification method that allows to measure the levels of DNA present in the biological sample of interest in real time. Amplification is monitored by using fluorescent DNA probes or intercalators, such as SYBR Green. This technique can be used to evaluate the mitochondrial genome (mtDNA), which, in the eukaryotic cell, is present in multiple copies. In this study, qPCR was used to observe and quantify how the level of mitochondrial DNA (mtDNA) varies in Human Embryonic Kidney (HEK 293) cells in which oxidative stress was induced by treatment with hydrogen peroxide (H2O2 ). The data obtained were normalized related to the amount of nuclear DNA and analyzed to understand how the level of mtDNA changes under these conditions. It was observed that treatment with H2O2 induced an increase in mitochondrial DNA in cells treated for 10 and 30 minutes, while the results suggested a decrease in mtDNA in the sample treated for 3 hours. The results obtained were discussed on the basis of the data present in the literature.
La tecnica della Quantitative Real Time PCR (qPCR) è un metodo di amplificazione del DNA che permette di misurare in tempo reale i livelli di DNA presenti nel campione biologico di interesse. L’amplificazione è monitorata mediante l’utilizzo di sonde o intercalanti fluorescenti del DNA, quali il SYBR Green. Questa tecnica può essere utilizzata per valutare il genoma mitocondriale (mtDNA), che, nella cellula eucariote, è presente in copie multiple. In questo studio, la qPCR è stata utilizzata per osservare e quantificare come varia il livello di DNA mitocondriale (mtDNA) in cellule Human Embrionic Kidney (HEK 293) nella quali è stato indotto uno stress ossidativo mediante trattamento con perossido di idrogeno (H2O2). I dati ottenuti sono stati normalizzati rispetto alla quantità di DNA nucleare e analizzati per capire come varia il livello di mtDNA in queste condizioni. Si è osservato che il trattamento con H2O2 induce un aumento del DNA mitocondriale nelle cellule trattate per 10 e 30 minuti, mentre i risultati suggeriscono una diminuzione di mtDNA nel campione trattato per 3 ore. I risultati ottenuti sono stati discussi sulla base dei dati presenti in letteratura.
