This paper analyses a selection of scientific articles concerning studies on sleep regulation in the model organism Drosophila melanogaster, with particular attention to the neuronal structures involved and the methodological approaches used. Sleep in this species, is controlled by genetic and neural mechanisms very similar to thos in mammals. In particular, the project focuses on the neurons of the dorsal fan-shaped body (dFB) and addresses the debate regarding the possible involvement of the ventral nerve cord (VNC), suggesting that sleep regulation is distributed across different areas of the nervous system. The study is based mainly of three key works: - Genetics of Sleep and Sleep Disorders, which emphasises the importance of Drosophila as an experimental model in the study of the genetic mechanisms underlying sleep; - Re-examining the role of the dorsal fan-shaped body in promoting sleep in Drosophila, which questions the centrality of the dFB by proposing a synergistic role for multiple neuronal regions; - Protocol for multiplexed RNAscope-based imaging of mRNAs in whole-mount adult Drosophila brains, which describes one of the main methodologies used for analysing the gene expression of neurons involved in sleep regulation.
Il presente elaborato analizza una selezione di articoli scientifici riguardanti gli studi sulla regolazione del sonno nell'organismo modello Drosophila melanogaster, con particolare attenzione alle strutture neuronali coinvolte e agli approcci metodologici utilizzati. Il sonno, in questa specie, è controllato da meccanismi genetici e neurali molto simili a quelli dei mammiferi. In particolare, il lavoro si focalizza sui neuroni del dorsal fan-shaped body (dFB) e affronta il dibattito riguardo il possibile coinvolgimento del midollo nervoso ventrale (VNC), suggerendo che la regolazione del sonno sia distribuita in diverse aree del sistema nervoso. Lo studio si basa principalmente su tre lavori chiave: - Genetics of Sleep and Sleep disorders, che sottolinea l’importanza di Drosophila come modello sperimentale nello studio dei mecanismi genetici alla base del sonno; - Re-examining the role of the dorsal fan-shaped body in promoting sleep in Drosophila, che mette in discussione la centralità di dFB proponendo un ruolo sinergico di più regioni neuronali; - Protocol for multiplexed RNAscope-based imaging of mRNAs in whole-mount adult Drosophila brains, che descrive una delle metodologie principalmente utilizzate per l’analisi dell’espressione genica dei neuroni implicati nella regolazione del sonno.
