This thesis examines the cortisol molecule, focusing on its synthesis, enzymatic inhibition, and analytical determination methods. The synthesis of cortisone following Horiguchi’s approach and the enantioselective reduction of cortisol mediated by the enzyme 11β-HSD1 are described. Several natural and synthetic inhibitors of 11β-HSD1 are also discussed as potential strategies to reduce cortisol levels and treat metabolic disorders. Finally, various analytical techniques for short- and long-term monitoring of cortisol concentration in the human body are presented.
La tesi analizza la molecola del cortisolo, approfondendone la sintesi, l’inibizione enzimatica e i metodi di determinazione. Viene descritta la sintesi del cortisone secondo Horiguchi e la sua riduzione enantioselettiva, mediata dall’enzima 11β-HSD1, per la sintesi del cortisolo. Sono inoltre esaminati diversi inibitori, naturali e sintetici, di 11β-HSD1 come possibile approccio per ridurre i livelli di cortisolo e trattare patologie metaboliche. Infine, vengono presentati vari metodi analitici per il monitoraggio a breve e lungo termine della concentrazione di cortisolo nell’organismo.
