This thesis investigates floral induction in apple (Malus × domestica Borkh.), the physiological process through which buds acquire a reproductive fate, thereby influencing flowering and the productive potential of the following season. In an apple-growing sector characterised by increasing varietal diversification, identifying the temporal window of this process may help explain differences in return bloom and responses to crop load, providing useful information for more targeted management of fruit thinning and vegetative-reproductive balance. The experiment was conducted at the Laimburg Research Centre on five cultivars of agronomic and commercial interest: Fuji, Golden Delicious, Gala, Red Delicious and Granny Smith. Buds collected from current-season shoots were sampled at eight stages between 19 and 143 days after full bloom (DAFB). The expression of MdAP1, MdFT1, MdFLC1c, MdbHLH48, MdGASA11/25 and MdBLH14 was analysed by qRT-PCR, using MdH3 as the reference gene, with the aim of identifying possible early or complementary molecular markers of the floral transition and comparing their temporal dynamics among cultivars. MdAP1 showed very low expression during the early sampling stages and increased mainly during the middle and late phases, a pattern consistent with its role in the consolidation of floral meristem identity rather than in the initial triggering of the transition. MdFT1, a flowering-promoting gene, showed changes from the early stages, but its expression profiles were not consistent among cultivars; the potentially transient nature of its expression and the relatively wide intervals between some sampling dates prevented the identification of a single, reliable temporal window of floral induction. MdFLC1c showed a peak at approximately 60 DAFB in several cultivars followed by a marked decline, consistent with an attenuation of the repressive component of flowering, although the influence of common seasonal factors cannot be excluded. MdbHLH48, analysed in Fuji, Golden Delicious and Gala, displayed a shared increase at 60 DAFB followed by cultivar-specific responses, consistent with a possible role in regulating the balance between MdFT1 and MdTFL1. MdGASA11/25 and MdBLH14 mainly showed non-synchronised increases during the middle and late stages and were therefore more informative about hormonal and metabolic regulation, particularly gibberellin signalling, than about the precise onset of floral induction. Overall, none of the genes analysed individually allowed the floral induction window of the five cultivars to be clearly and generally defined. Nevertheless, the results indicate that a relevant reorganisation of the bud regulatory network occurs around 60 DAFB, with simultaneous modulation of flowering-promoting and flowering-repressing signals. The main limitations include the presence of buds at different developmental stages within the samples, the intervals between some sampling dates, and the absence of histological observations and direct measurements of crop load and phytohormone levels. The results therefore support an integrated approach based on multiple genes with complementary functions, combined with morphological characterisation of buds, quantification of phytohormones and crop load, and more frequent sampling. Such an approach could help identify more reliable indicators of the floral transition and, in the longer term, support more precise management of return bloom and crop load in multi-cultivar apple orchards.

Analysis of floral induction in different apple cultivars through gene expression profiling
