The work carried out during the internship aims to analyze the effect of polyethylene terephthalate nanoparticles (nPET) on the cytotoxic immune responses of the colonial ascidian Botryllus schlosseri, a model organism belonging to the Tunicata group, the sister group of vertebrates. To pursue this goal, degranulation assays were performed on hemocytes in vitro to evaluate the cytotoxic immune response after exposure to nanoplastics. To support the results obtained, the activity of phenol oxidase, an enzyme contained in these cells, was evaluated through spectrophotometric analysis.
Il lavoro svolto durante il periodo di internato si pone l’obbiettivo di analizzare l’effetto delle nanoparticelle di polietilene tereftalato (nPET) sulle risposte immunitarie citotossiche dell’ascidia coloniale Botryllus schlosseri, un organismo modello appartenente al gruppo dei Tunicata, sister group dei vertebrata. Per perseguire tale scopo sono stati effettuati su emociti dei saggi di degranulazione in vitro, al fine di valutare la risposta immunitaria citotossica dopo l’esposizione alle nanoplastiche. Per avvalorare i risultati ottenuti é stata valutata l’attività della fenolo ossidasi, un enzima contenuto in queste cellule, tramite analisi spettrofotometriche.
