In this thesis, plants of Vitis vinifera cv Chardonnay edited by Genome-Editing have been obtained and characterized. Specifically, this editing involves the silencing of susceptibility genes against Erysiphe Necator (etiological agent of the Oidium) and various biotrophic fungi. The inactivation of these genes cause the non-recognition of the plant by the pathogen that induce a resistance mechanism called Broad-Spectrum Resistance (BSR). The analyzes were made with the use of Real Time PCR for the evaluation and quantification of the number of T-DNA copies in the plant genome. Following the preparation of the samples for Illumina MiSeq sequencing, the results were analized to identify the changes obtained in the plant genome
Nella presente tesi sono state ottenute e caratterizzate piante di Vitis vinifera cv Chardonnay editate mediante Genome Editing. Nello specifico tale editing prevede il silenziamento di geni di suscettibilità nei confronti di Erysiphe Necator (agente eziologico dell’Oidio) e nei confronti di diversi funghi biotrofi. L’inattivazione di tali geni di suscettibilità determina il non riconoscimento della pianta da parte del patogeno, attivando un meccanismo di resistenza definito Broad-Spectrum Resistance (BSR). Le analisi svolte prevedono l’uso della Real Time PCR per la valutazione e quantificazione del numero di copie di T-DNA presente nel genoma della pianta. In seguito alla preparazione dei campioni per il sequenziamento Illumina MiSeq sono stati analizzati i risultati per l’identificazione delle modifiche ottenute all’interno del genoma della pianta.
