Donkey milk is a healthy source of nutrition and its benefits have aroused great interest among the scientific community over the last few years, this is due to the fact that it has both properties similar to those contained in breast milk, and it is characterized by low allergenicity compared to cow’s milk. Indeed, the development of fermented milks and fermented dairy products has become a case of study that aims to meet and satisfy the customers’ needs. There is no specific law governing donkey milk production methods and trade, nevertheless reference is made to Regulation (EC) N° 1662/2006 which grants its sale permission, according to this specific clause, which concerns “other species”, the sale of raw donkey milk is only allowed whether the total microbial count of the sample is lower than 1.500.000 CFU/mL at 30°C. However, there are some studies in literature where potentially pathogenic micro-organisms have been detected. Therefore, the aim of this thesis is to demonstrate the applicability of analytical methods to complex dairy matrices, such as donkey milk. Samples of donkey milk, supplied by two different companies in Northern Italy, were tested using first the Real time PCR (direct method) and then the Elisa Test (indirect method) in order to detect the potential presence of Coxiella burnetii, the bacteria responsible for Q Fever. The survey was conducted at the “Istituto Zooprofilattico Sperimentale delle Venezie” (PD). In conclusion, the results showed that these methods can be applied to the donkey milk matrix. Tests have also detected the presence of Coxiella burnetii antibodies and DNA of Coxiella burnetii through Real Time PCR, an outcome that suggests the need to establish a monitoring plan to better monitor the donkey milk production.

Il latte d’asina è un alimento che negli ultimi anni sta destando particolare interesse nella comunità scientifica, sia per le sue proprietà molto simili al latte materno, ma anche per la bassa allergenicità rispetto al latte vaccino. Si sta studiando, infatti, lo sviluppo di latti fermentati e altri prodotti a base di questo latte che soddisfino le esigenze dei consumatori. A livello legislativo, tuttavia, non è presente una normativa specifica riguardante la produzione e commercializzazione del latte d’asina, ma si fa riferimento al Regolamento (CE) 1662/2006, secondo il quale è permessa la vendita di latte d’asina crudo sotto la clausola “Altre specie produttrici di latte” solo se la carica microbica totale risulta essere inferiore a 1.500.000 CFU/mL a 30°C. In letteratura sono presenti, tuttavia, studi dove sono stati rilevati microrganismi potenzialmente patogeni al suo interno. Questo lavoro vuole dimostrare la possibile applicabilità di un metodo d’analisi alla matrice latte d’asina; per dimostrare ciò, sono stati testati dei campioni di latte d’asina crudo, acquistati in due aziende differenti del Nord Italia, attraverso le metodiche d’analisi Real-Time PCR (metodo diretto) e Test ELISA (metodo indiretto), per rilevare l’eventuale presenza di Coxiella burnetii, un microrganismo potenzialmente patogeno e responsabile della Febbre Q. Le analisi sono state condotte presso i laboratori dell’Istituto Zooprofilattico Sperimentale delle Venezie (PD). Dai risultati ottenuti in questo lavoro, il metodo risulta applicabile a questa matrice, inoltre sono emerse delle positività sia per quanto riguarda la presenza di anticorpi contro Coxiella burnetii nel latte, ma anche la positività di un campione alla Real time PCR, ciò suggerisce la necessità di istituire un piano di monitoraggio lungo la filiera di questo prodotto.
